I think this is a tricky question because some people have a very different sense of humor. For me, the funniest books are the ones written by the actor/comedian Steve Martin. His books are some of the few that I've actually laughed out loud while reading.

In following the conquests of the Man of LaMancha, Don Quixote, I roared with laughter. Cervantes had me giggling, smiling, and laughing to the point of tears. It was embarrassing.

Ditto from DATo !!! It is hard to believe that this book was written 500 years ago. Cervantes humor is timeless. I too rolled with laughter many times while reading this book, a few times nearly from the floor.
